Having painted panelling numerous times we always give it a light sanding before the primer. 80 or 100 grit and do it by hand not an electric sander. A drywall pole sander works great then wipe down. It's a bit of extra work but the paint sticks better.

Good tips here for painting paneling.  Many people assume it's not possible, but we've had clients do it successfully and it really updates the look of the home when the "knots" are hidden!
